Am married to a US citizen, and am about to apply for my I-130 and I-485 concurrently, my wife has been working but she never file her tax returns for 2012 and 2013, all I have is her W2 forms. Will that be a problem?

Yes, it will be a problem. File the 2013 tax return, using her W2's, so that you won't receive a Request for Evidence and have your processing held up until she provides a copy of the most recent tax year tax return.
