Canadian Green Card Holder in US -- Wife has international opportunity

I'm a green card holder (spose visa) living in the US. We own a home here and my name is on the title.
My wife has a unique opportunity to work in the U.K. for an American company. Of course I'll accompany her and hopefully find work there. How would this affect my green card status? Would being a home owner provide me any leverage?

Depends on the time spent outside the country. It will eventually affect your citizenship as you need to have met the residency requirements before filing

You can safely stay outside the country for upto 2yrs on your greencard, if you file for a Re-Entry permit while in the US.
