Hi,
I am a US citizen and I would like to file form I-130 for my mother, brother and 2 sisters at the same time, do I have to file affidavit of support form I-864 now with I-130 or I have to wait until USCIS approve their application?

I-864

You don't have to file I-864 yet. You have to wait for your I-130 to get approved and then your case will be moved to NVC and in case of your parents, there visa category will be current since it is unlimited (till the new immigration bill passes) but for your brother(s)/sister(s), you will have to wait till there visa category becomes current and in both these cases - parents and siblings - NVC will send you a packet including I-864 form which you will have then have to fill up per the instructions.
