the mother is not your stepdaughter's "husband/wife" and the sister is not your stepdaughter's child, right? then they do not get written down.

On another note, I saw in another post that you are filing an I-130 for your wife and her two children. I assume the stepdaughter in your question above is your wife's daughter. On the I-130, your wife is the Relative in part C, you are the Relative's "husband" in question 17 and the two children are the Relative's "children" in question 17.

Whether you need to file two more I-130, one for each of the children, (Praetorian, is that true?), I don't know, but if you do, then each of the children are the Relatives in part C on their respective I-130, and they don't have any "husbands/wives" or "children" on question 17 (I assume that is the case).
