My wifes bro is in US with GreenCard from 5 years + married to a US Citizen since then.

What is the best way to immigrate for his married sister + family? is F4?
Or first sponsor his parents to US and then the parents sponsor their daughter?

Yes, the best way is to do family immigration through F4 category after he is US citizen.

After he is US citizen, he could sponsor his parents and after parents become citizens, they could sponsor the daughter. However, with that route, you really save less than 1 year, may be few months. And parents must be able to become citizens, such as know English and be able to pass the test etc.

What if he (USC) applies for his sister and family, how many years that process would take? Oh, english test is not a problem.

Directly sister - Around 13 years.
First parents and then their daughter - Over 12 years.
