I am petitioning my 45 yrs old brother to US and my wife’s cousin is asking me if I can co-sponsor her and husband since his adult son can’t comply to the income requirement. If I say yes will this affect my brother’s impeding petition?

It depends, do you have enough income to sponsor 3 persons given your existing household size? If yes, then your brother's petition will not be affected. If no, it may be.

Yes, both beneficiaries are getting an immigrant visa and therefore need to be sponsored. If you uptake their responsibility, you will add them each to your "household size".
