Form I-130 file for brother

If I file I-130 for my brother do I have to file a separate I-130 for his wife?

No. You can't (you can't petition any "-in-law" relatives), and you don't need to (spouse and unmarried under-21 children that the principal beneficiary has at the time he immigrates can immigrate as derivative beneficiaries).
