My application for my brother which was submited back in 2002 is approved & is waiting for a visa. my brother just got married 5 months ago. Do i need to updated my application to include his wife? and if yes, how do i do it? does this affect the visa proccessing time???

You don't have to worry about anything. Once his PD is current, he can add his wife as Derivative beneficiary and she will also get a Green Card. His Priority date should go current anytime soon, you need to check the visa bulletin.
