Chances of Tier 1 Visa

Hi Friends,

I am going to apply for Tier 1 Visa (General) category.
After checking with points based calculator, I have enough score to qualify for this visa.

I have tried for US Visiting Visa (Business) through my previous employer in year 2005, but it was refused since I was single and not visited any country before.

But after that I have visited Italy & Dubai on Business Trip.

Is there any chance that my UK visa will be given?
Do they take my US visa refusal in to account?
