Hi guys!
It has been couple weeks now I've submitted a I-129F form for my fiancee and our 3.5-year-old daughter. I would like to know if a DNA test will be a requirement once the petition is approved?

There's some mismatch here. You say K3 visa? do you mean K-1 and K-2 for your fiance and her child? K-3 is for married couples...
Also, if you're filing based on a fiance visa, then a DNA test is not required to show parentage between beneficiary (I'm assuming that's what you meant).
If you're filing based on marriage (k-3 and k-4 visa) then you may have to prove parentage through a DNA test. But that's up to the USCIS or US Consulate to ask.. you cannot do it without their request.
Final note: K-3 and K-4 are outdated and obsolete. You will either file a fiance visa K-1 and K-2 for unmarried couples. Or you file the i-130 for a cr1/ir1 for married couples.

Thanks a lot for the answer. Yes, I meant K1 and K2 visa. I'm also confused about the intent to marry letter. I've submitted one for myself along with the form I-129f. Should I also join my fiancee's letter or would she have to provide it once she'd be called for the Interview at the embassy?

For the K-1 and K-2 visa process, there's no DNA test required from you to compare to the beneficiary or her children.
The letter of intent for the I-129F has to be signed by BOTH you and your fiance. If you submitted it for yourself but not hers, then the USCIS is going to ask you to submit hers, too. The K visa interview doesn't require a letter of intent to marry.
