Hi,
I have applied labor through employer A in oct 2008. 3 months later they said that it has gone to audit. At present Iam with employer B. My question is
1. Can I use my previous labor(which is from A) eventhough Iam with employer B and extend my stay in US?
2. Do I have to apply labor through employer B to extend my stay in US?

If with A, it is in audit, then you can't.
2. Do I have to apply labor through employer B to extend my stay in US?
Yes.

Here, if you plan to get extension with A, then A should provide valid docs of project and everything for you, on which you will be working.
