I have LCA for Location X, which does not has any direct flights. I would like to enter USA from Location Y. After a break of week - 10 days I would head to Location X (my LCA location). Would this cause any problem?

Is it mandatory to directly go to LCA location?

I don't think so. This seems very very unlikely. Bottom line they will ask you why you come through that airport and you can explain why.
