I would like to start by saying I live in Jakarta, Indonesia, I am a US Citizen (by birth), and my wife is Indonesian. My wife was denied a B2 visa two days ago on the basis of 214b. So of course we are going to reapply. Since I am the one who will be financially responsible for her during the trip, should I (can I) file an I-134?

You can provide an I-134 with supporting documents to demonstrate your financial ability to provide for her travel expenses. But you can't really sponsor your wife (or anyone) for a B2 visa.
