Single entry reapplier

If one is on a single entry 6 month B2 visa and is a doctor, on returning to his / her home country and reapplying, will he get rejected?

If you have not overstayed your first visit and your purpose of visit on the next application is concrete, the chances of getting the visa are good.
Please elaborate why and when you wish to apply the second time for better advice. What was your purpose the first time? Also state whether they put an annotation( a note restricting your visit for a specific purpose or place) on your 6 month visa and what was it?
If you are still in US on your first visa and plan on settling in US permanently, please state.
