Ds-160 question any relative in united state

i am u.s. Citizen. I want to apply for tourist visa for one of my brother who is in india. I got another brother who is in united state but he is living illegally but i filed his petition i-130 form for him.now my question is
in the ds-160 form it ask do you have any relative in united state does he needs to say yes or no to that question.
