my friends works in singapore and he wants to come to US in visitor visa. Does he needs to apply from india only or he can apply from singapore. he is neither a Permanent resident or a citizen of singapore.

*Is he married? If yes, are wife (kids) in singapore or in India?
*What are his family or other ties in India?

Should apply in singapore since he is developing permanent ties here.
Since he is young and single, it may not be easy to get the visa if his purpose of visit is very
casual like visiting a friend or sightseeing.

He must show purpose of visit that seems more natural like visiting a close relative in US for 2-3 weeks vacation Or going on behalf of his company for a business trip Or
if he was married, he could show a sighteeing trip to US with his family .
